Abstract
AccordingtoreportfromGlobalCancerObservatory(GLOBOCAN),cervicalcancerisratedas oneofthemostdominanttypesofcancerinNigeria.Traditionally,medicinalplantsareknown tobewellutilizedintreatingcancerousdiseases.Theresearchreportedhereinthereforeaimedat screeningtheextractsofCitrulluscolocynthis(CC)andTalinum fruticosum (TF)fortheir potentialcytotoxiceffectsagainsttargetedcancercells.Qualitativephytochemicalscreeningof thesecondarymetaboliteswasdone.GC-MSanalysisofTFextractwasengagedtoevaluatethe compoundsforstructure-activityrelationship(SAR).Twelvecompoundswereselectedfor moleculardocking.TheHeLacellline,amodelofcervicalcancer,wasthenexposedfor24h then48htoarangeofconcentrations,upto500μg/ml,oftheethylacetate(CCE)andmethanol extracts(CCM)ofthewholefruitofC.colocynthis,followedbycellviabilityquantificationwith the [3-(4,5-dimethylthiazol-2-yl)-2,5-dipenyltetrazolium bromide] (MTT) assay. The phytochemicalscreeningconfirmedphenols,tannins,flavonoids,cardiacglycosides,saponins, triterpenoidsandalkaloidsinthesamplevegetables.MoleculardockingstudyrevealedTalinum fruticosum topossesshighbindingaffinityforprostate,breastandcervicalcancertreatment throughandrogenreceptor,profiling-1andHPV51E-6inhibition.Thepreliminarycytotoxicity assayat24hshowedCCEwasthricemorepotentlycytotoxicwithanIC50valueof33.95±3.39 (μg/ml)thanCCM withanIC50valueof101.59±24.37.TheIC50(μg/ml)valueforthe48h treatmentwasfoundtobe16.0±5.7forCCEand76.3±12.9forCCM (p<0.05).FT-IR analysisshowedimportantfunctionalgroupssuchasthehydroxyl,alkane,amine,amideand acidfunctionalities.Thefindingstherefore,suggestthatCitrulluscolocynthisandTalinum fruticosumcouldserveasapotentialdrugforprostate,breastandcervical.
